چکیده

Histological staining of wild-type and sevenless transgenic Drosophila melanogaster bearing Rh3lacZ fusion genes permits the selective visualization of polarization-sensitive R7 and R8 photoreceptor cells located along the dorsal anterior eye margin. Diffusion of fl-galactosidase throughout these cells reveals that they project long axons to the two most peripheral synaptic target rows of the dorsal posterior medulla, defining a specialized marginal zone of this optic lobe. Comparison of the staining patterns of marginal and nonmarginal Rh3-lacZ-expressing photoreceptor cells in the same histological preparations suggests that the marginal cells possess morphologically specialized axons and synaptic terminals. These findings are discussed with reference to the neuroanatomy of the corresponding dorsal marginal eye and optic lobe regions of the larger dipterans Musca and Calliphora, and in relation to the ability of Drosophila to orient to polarized light.

A versatile maze for learning and geotaxic selection in <Emphasis Type="Italic">Drosophila melanogaster </Emphasis>

Behavior geneticists have long sought to display learning in the fruit fly (Drosophila melanogaster), but previous research efforts have proven problematic. The following describes a maze apparatus and a procedural technique that have produced positive results in learning tasks with negative geotaxis flies. The Plexiglas maze is inexpensive to construct and offers a functional versatility that ...

Spatial and temporal expression of the period gene in Drosophila melanogaster.

The temporal and spatial expression of the period gene of Drosophila melanogaster has been analyzed by examining the expression of a per beta-galactosidase fusion gene in transformants and by in situ hybridization experiments with wild-type flies. Several strains of Drosophila melanogaster, transformed with the fusion gene, have been generated. The gene is active in mid-late embryos in the midl...

bantam Is Required for Optic Lobe Development and Glial Cell Proliferation

microRNAs (miRNAs) are small, conserved, non-coding RNAs that contribute to the control of many different cellular processes, including cell fate specification and growth control. Drosophila bantam, a conserved miRNA, is involved in several functions, such as stimulating proliferation and inhibiting apoptosis in the wing disc. Here, we reported the detailed expression pattern of bantam in the d...

Generation and early differentiation of glial cells in the first optic ganglion of Drosophila melanogaster.

We have examined the generation and development of glial cells in the first optic ganglion, the lamina, of Drosophila melanogaster. Previous work has shown that the growth of retinal axons into the developing optic lobes induces the terminal cell divisions that generate the lamina monopolar neurons. We investigated whether photoreceptor ingrowth also influences the development of lamina glial c...

A combined molecular and cytogenetic approach to genome evolution in <Emphasis Type="Italic">Drosophila</Emphasis> using large-fragment DNA cloning

Methods of genome analysis, including the cloning and manipulation of large fragments of DNA, have opened new strategies for uniting molecular evolutionary genetics with chromosome evolution. We have begun the development of a physical map of the genome of Drosophila virilis based on large DNA fragments cloned in bacteriophage P1. A library of 10,080 Pl clones with average insert sizes of 65.8 ...
